產(chǎn)品概述

干擾素γ（IFN-r，也稱為II型干擾素）原本被認(rèn)定為是一種抗病毒活性所產(chǎn)生的促有絲分裂原激活T淋巴細(xì)胞。蛋白質(zhì)的組成與干擾素β或各種干擾素α家族蛋白沒有顯著的同源性。干擾素γ主要通過T淋巴細(xì)胞和自然殺傷細(xì)胞產(chǎn)生。干擾素γ的生產(chǎn)是由抗原的刺激和細(xì)胞因子引起，如IL-12（白細(xì)胞介素12）。小鼠的干擾素γ編碼155個(gè)氨基酸殘基的前體蛋白與疏水信號肽，后者是裂解出的133氨基酸殘基的成熟蛋白。在溶液中，干擾素γ顯示存在一個(gè)非共價(jià)連接的同型二聚體與IL-10（白細(xì)胞介素10）極為相似。鼠的干擾素γ顯示，大約40 ％氨基酸殘基與人的干擾素γ同源，沒有種屬的交叉反應(yīng)。

產(chǎn)品原理

本實(shí)驗(yàn)采用雙抗體夾心ELISA法?？剐∈驣FN-γ單抗包被于酶標(biāo)板上，同時(shí)加入標(biāo)本、標(biāo)準(zhǔn)品和生物素化的抗小鼠IFN-γ抗體，標(biāo)本、標(biāo)準(zhǔn)品中的IFN-γ會(huì)與加入于孔中的生物素化的抗小鼠IFN-γ抗體及包被于酶標(biāo)板上的單抗結(jié)合，形成免疫復(fù)合物，游離的成分被洗去；加入辣根過氧化物酶標(biāo)記的親合素，親合素與生物素特異性結(jié)合，游離的成分被洗去。加入顯色底物（顯色劑），若反應(yīng)孔中有IFN-γ，辣根過氧化物酶會(huì)使無色的顯色劑變藍(lán)，加終止液變黃。在450nm處測OD值，IFN-γ濃度與OD₄₅₀值之間呈正相關(guān)，可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中IFN-γ濃度。

產(chǎn)品性能

1. 靈敏度：最小可測小鼠IFN-γ達(dá)4 pg/ml。

2.重復(fù)性：板內(nèi)、板間變異系數(shù)均<10%。

產(chǎn)品組分及儲(chǔ)存條件

試驗(yàn)所需自備儀器和試劑:

1. 酶標(biāo)儀(450nm檢測波長濾光片，570nm或630nm校正波長濾光片)。

2. 高精度加液器及一次性吸頭：0.5-10μl，2-20μl，20-200μl，200-1000μl。

3. 37℃恒溫箱、蒸餾水或去離子水、坐標(biāo)紙。
